测定方法

---

1．称取3g硫酸铜(CuSO₄)加入到30ml普通氢氧化氨(NH₄OH)溶液中，放入有盖玻瓶内。

2．两种物质混合后，会形成一种浅兰色的沉淀物(Cu(OH)₂)。如果没有沉淀形成，需再加入CuSO₄，直至出现沉淀为止。因为普通的NH₄OH溶液的浓度会有差异。这样做可保证使CuSO₄与NH₄OH之间能完全反应。但须注意，反应过剩的氨气对人的眼睛有刺激作用。

3．刺破甜三叶种子的种皮，以便充分吸胀。

4．将种子放在玻璃容器里的水中，让其吸胀2－5小时。

5．当种子吸胀后，沥去多余水分，加入足够的CuSO₄-NH₄OH混合液淹没种子。

6．经染色1－10分钟后，黄色甜三叶种子的种皮被染成深棕色至黑色；而白色甜三叶种皮被染成淡绿色或黄绿色。
